目的鉴定分离自龙岗区的金黄色葡萄球菌,探讨使用金黄色葡萄球菌spa基因对菌株进行多态性分析的价值。方法在Gen Bank数据库中查找金黄色葡萄球菌spa基因序列,并设计引物进行实时荧光PCR技术分型,建立新的金黄色葡萄球菌分型方法,并与常规金黄色葡萄球菌spa基因分型和脉冲场凝胶电泳(PFGE)分型法进行比较,构建龙岗区金黄色葡萄球菌分型数据库。结果 5例患者抗结核治疗前后前鼻腔定植的金黄色葡萄球菌spa分型相同,结果与PFGE结果一致;5例患者定植的金黄色葡萄球菌spa分型分别为:t795、t179、t189、t758和t796,其中t795、t758和t796为新发现的分型。结论 spa分型是一个较好的金黄色葡萄球菌快速基因分型方法。
Objective To identify Staphylococcus aureus isolated from Longgang District and explore the value of using Staphylococcus aureus spa gene to analyze the polymorphism of the strains. Methods The gene sequences of spa of Staphylococcus aureus were searched in Gen Bank database and primers were designed for real-time fluorescence PCR. A new typing method of Staphylococcus aureus was established and compared with the conventional genotyping and pulse of Staphylococcus aureus Field gel electrophoresis (PFGE) typing comparison, build Longgang District Staphylococcus aureus typing database. Results The spa type of Staphylococcus aureus colonized by the nasal cavity before and after anti-TB treatment in 5 patients was the same. The results were consistent with those of PFGE. The spa genotypes of Staphylococcus aureus colonized in 5 patients were t795, t179, t189, t758 and t796, of which t795, t758 and t796 are newly found subtypes. Conclusion Spa typing is a good method for rapid genotyping of Staphylococcus aureus.
