目的 :探讨人早幼粒细胞白血病细胞内STAT3的表达及酪氨酸磷酸化活化情况。方法 :培养人早幼粒细胞白血病细胞株HL 60 ,利用特异性抗体 ,用免疫沉淀法、聚丙烯酰胺凝胶 (SDS PAGE)电泳及WesternBlot方法进行检测。结果 :实验结果表明HL 60细胞内有STAT3α和STAT3 β的表达和酪氨酸 (Y70 5 )磷酸化活化。 结论 :本研究结果提示STAT3的异常表达和活化可能与人早幼粒细胞白血病的发生有关。
Objective: To investigate the expression of STAT3 and tyrosine phosphorylation in human promyelocytic leukemia cells. Methods: Human promyelocytic leukemia cell line HL 60 was cultured and detected by immunoprecipitation, polyacrylamide gel electrophoresis (SDS PAGE) and Western Blot using specific antibodies. Results: The results showed that the expression of STAT3α and STAT3β and the activation of tyrosine (Y70 5) phosphorylation in HL 60 cells. Conclusion: Our results suggest that aberrant STAT3 expression and activation may be related to the occurrence of human promyelocytic leukemia.
