牛冠状病毒BCV-Aks-01新疆南疆株全基因组序列测定及其分子特征分析

为掌握牛冠状病毒(Bovine coronavirus,BCoV)BCV-Aks-01新疆南疆株的全基因组序列、分子结构特征及遗传变异情况。本研究以新疆南疆某牛场BCV-Aks-01阳性样本为基础,以GenBank中BCoV参考株设计扩增引物和测序引物,提取样本中病毒核酸,对BCV-Aks-01株进行全基因组扩增、测序、构建系统发生树及序列分析。结果表明,BCV-Aks-01新疆南疆株为BCoV,其全基因组长为30 975bp,与GenBank收录的参考株比较,核苷酸同源性高达98.8%,属于β类冠状病毒2a亚类,具有β类冠状病毒共同结构特征。首次获得BCV-Aks-01新疆南疆株全基因组序列并阐明了其基因组分子结构特征。 In order to understand the whole genome sequence, molecular structure and genetic variation of BCV-Aks-01 Xinjiang southern Xinjiang strain of Bovine coronavirus (BCoV). In this study, based on BCV-Aks-01 positive samples from a cow farm in southern Xinjiang, the BCV-Aks-01 strain of BCV-Aks-01 strain was amplified by PCR using the BCoV reference strain in GenBank. Increase, sequence, construct phylogenetic tree and sequence analysis. The results showed that the BCV-Aks-01 southern Xinjiang Xinjiang strain was BCoV, with a total genome length of 30 975 bp. Compared with the reference strain contained in GenBank, the nucleotide sequence of BCoV was as high as 98.8% With the coronavirus beta common structural features. The whole genome sequence of BCV-Aks-01 southern Xinjiang was obtained for the first time and its molecular structural characteristics were elucidated.
