目的研究宫颈癌组织中自噬基因ARHI与Beclin1蛋白的表达。方法采用免疫组化方法对非宫颈病变12例、宫颈上皮内瘤变(CINⅡ~Ⅲ)15例、宫颈癌50例(无淋巴结转移患者34例,淋巴结转移患者16例)进行试验,检测各组织中ARHI、Beclin1蛋白的表达。结果 (1)ARHI与Beclin1在正常宫颈组、宫颈上皮内瘤变组及宫颈癌组中的表达依次下降,分别比较两者的三组间差异,均有统计学意义(P<0.05);(2)ARHI的表达与宫颈癌临床分期、分化程度有关,差异有显著性(P<0.05);(3)ARHI与Beclin1在宫颈癌组织中的表达具有正相关性(r=0.387,P=0.006)。结论随着宫颈病变的加重,ARHI及Beclin1的表达均逐渐降低,且在宫颈癌组织中二者的表达具有正相关性,提示二者可能共同参与了宫颈癌的发生,其表达的下降可能与宫颈病变的进展有关。
Objective To study the expression of autophagy genes ARHI and Beclin1 in cervical cancer. Methods Immunohistochemical method was used to test 12 cases of non-cervical lesions, 15 cases of cervical intraepithelial neoplasia (CINⅡ-Ⅲ), 50 cases of cervical cancer (34 cases without lymph node metastasis and 16 cases with lymph node metastasis) In ARHI, Beclin1 protein expression. Results (1) The expression of ARHI and Beclin1 in normal cervical cancer group, cervical intraepithelial neoplasia group and cervical cancer group decreased in turn, and the differences between the three groups were statistically significant (P <0.05); ( The expression of ARHI was positively correlated with the clinical stage and differentiation of cervical cancer (P <0.05). (3) There was a positive correlation between the expression of ARHI and Beclin1 in cervical cancer (r = 0.387, P = 0.006 ). Conclusions With the increase of cervical lesions, the expression of ARHI and Beclin1 are gradually decreased, and there is a positive correlation between the expression of ARHI and Beclin1 in cervical cancer, suggesting that the two may participate in the carcinogenesis of cervical cancer.
