目的:观察加减知柏地黄丸对抗特发性中枢性性早熟(idiopathic central precociouspuberty,ICPP)的作用并初步探讨其作用机制。方法:采用瘦素(Leptin)诱导幼龄雌性BALB/c小鼠建立ICPP小鼠模型,以甲地孕酮为阳性对照药,并另设GnRH-A(抑那通)对照组,观察该中药复方对模型小鼠血清激素雌二醇(E2)和黄体生成素(LH)水平、阴道开口情况、阴道脱落细胞涂片、子宫、卵巢重量及体重、子宫指数和卵巢指数、子宫和卵巢组织病理学变化等指标的影响。结果:加减知柏地黄丸可抑制模型小鼠的阴道开口提前、子宫和卵巢重量增加、子宫指数和卵巢指数升高、及LH和E2水平的升高,实验中除GnRH-A组外,模型组和各给药组的阴道脱落细胞涂片、子宫、卵巢病理切片镜下所见均无明显差别。实验所见加减知柏地黄丸、甲地孕酮对各观察指标的影响之间差别无统计学意义。结论:加减知柏地黄丸对瘦素刺激诱导的ICPP模型具有一定的对抗作用,其机制可能是通过抑制瘦素诱导启动的垂体——性腺轴功能,抑制垂体分泌LH,进而抑制E2分泌,或直接抑制瘦素对性腺的作用。
Objective: To observe the effect of Jiabai Dihuang Pills against idiopathic central precocious puberty (ICPP) and to explore its mechanism. METHODS: Leptin was used to induce young female BALB/c mice to establish an ICPP mouse model. Megestrol was used as a positive control and a GnRH-A (Istatin) control group was established. The traditional Chinese medicine was observed. Compound model mouse serum hormones estradiol (E2) and luteinizing hormone (LH) levels, vaginal opening, vaginal exfoliated cell smears, uterus, ovary weight and body weight, uterine index and ovarian index, uterus and ovarian tissue disease Impact of indicators such as changes in science. RESULTS: The addition of Zhibai Dihuang Pill can inhibit the advancement of vaginal opening, the increase of uterus and ovary weight, the increase of uterine index and ovarian index, and the increase of LH and E2 levels in model mice except for GnRH-A group. There was no significant difference in the vaginal exfoliated cell smears, uterus, and ovary pathological sections of the model group and the respective administration groups. There was no statistically significant difference in the effect of addition and reduction of Zhibai Dihuang Wan and Megestrol on the observed indexes in the experiment. Conclusion: The addition and reduction of Zhibai Dihuang Pill have a certain antagonistic effect on the ICNP model induced by leptin stimulation. Its mechanism may be through inhibiting the pituitary-gonadal axis function initiated by leptin, inhibiting LH secretion from pituitary, and inhibiting E2 secretion. Or directly inhibit the effect of leptin on the gonads.
