利用RNAi和表达谱芯片分析日本血吸虫Sj34.9基因的功能

目的分析日本血吸虫新基因Sj34.9的功能,为进一步研究该基因提供依据。方法采用RNA干扰(RNAi)技术对Sj34.9基因在血吸虫体内的表达进行干扰,利用血吸虫寡聚核苷酸表达谱芯片分析Sj34.9基因表达沉默后血吸虫基因的表达情况。结果 Sj34.9基因RNAi组与对照组相比,显著差异表达的基因共378个,其中上调基因202个,下调基因176个;通路分析显示,表达差异基因主要与信号转导、信号分子间相互作用及各类物质代谢有关;基因本体论分类结果显示,多数基因与分子间结合、膜构成、细胞过程和机体生长发育及代谢相关。结论 Sj34.9基因可能对日本血吸虫的形成、生长、发育、调节、代谢等具有重要作用。 Objective To analyze the function of Sj34.9, a novel gene of Schistosoma japonicum, and provide the basis for further study of this gene. Methods RNA interference (RNAi) was used to interfere with the expression of Sj34.9 gene in Schistosoma japonicum, and the expression of Sj34.9 gene was analyzed by using Schistosoma japonicum oligodeoxynucleotide microarray. Results There were 378 genes that were significantly differentially expressed in Sj34.9 RNAi group compared with the control group, of which 202 genes were up-regulated and 176 genes were down-regulated. Pathway analysis showed that the differentially expressed genes were mainly associated with signal transduction and signaling molecules Role and various types of material metabolism; gene ontology classification results show that most genes and molecular binding, membrane formation, cellular processes and body growth and metabolism and related. Conclusion Sj34.9 gene may play an important role in the formation, growth, development, regulation and metabolism of Schistosoma japonicum.
