目的 :引入一种外源性雌激素的快速筛检方法 ,检测外源性化学物的雌激素样活性。方法 :用人乳腺癌细胞体外增殖法 ( E- SCREEN) ,检测硫丹等化学物的雌激素样活性。结果 :成功地建立了 E- SCREEN法 ,该法可检测到 1× 10 - 13 m ol/ L的雌二醇。发现在该试验条件下 ,己烯雌酚为雌激素受体的全激动剂 ,硫丹为雌激素受体的部分激动剂 ,而 4 -羟化他莫昔酚无雌激素样作用。结论 :E- SCREEN法敏感性高 ,并有快速、简便等优点 ,适用于大量化合物的初筛。
OBJECTIVE: To introduce a rapid screening method for exogenous estrogen to detect estrogen-like activity of exogenous chemicals. Methods: The estrogen-like activity of endosulfan and other chemicals was detected by the human breast cancer cell proliferation assay (E-SCREEN). RESULTS: The E-SCREEN method was successfully established, which detected 1 × 10-13 mol / L estradiol. Diethylstilbestrol was found to be a full agonist of the estrogen receptor under the experimental conditions. Endosulfan is a partial agonist of the estrogen receptor whereas 4-hydroxytamoxifen has no estrogen-like effect. Conclusion: The E-SCREEN method has high sensitivity and is fast and simple. It is suitable for screening a large number of compounds.
