作者用白蛋白包埋法制成具有磁响应的秋水仙碱-白蛋白-磁微粒,并采用MTT法检测了秋水仙碱-白蛋白-磁微粒及游离秋水仙碱对EMT_6细胞的生长抑制作用,对乳癌EMT_6细胞裸鼠移植瘤进行基底部注射和外加磁场定位治疗.结果表明:秋水仙碱-白蛋白-磁微粒及游离秋水仙碱在体内、体外实验均对肿瘤细胞有明显的杀伤作用,体外实验在100μg/ml剂量时,灭活率分别为56.16%和58.46%.对乳癌EMT_6细胞裸鼠移植瘤的抑瘤率分别为32.8%和23.6%,提示秋水仙碱-磁微粒外加磁场引导定位对裸鼠乳腺癌有较好的抑制作用.
The authors made a magnetically-responsive colchicine-albumin-magnetic microparticle by albumin embedding, and the inhibitory effects of colchicine-albumin-magnetic particles and free colchicine on the growth of EMT_6 cells were measured by MTT assay. Breast cancer EMT_6 cell xenografts in nude mice were treated by basal injection and external magnetic field. The results showed that colchicine-albumin-magnetic particles and free colchicine had obvious killing effect on tumor cells in vitro and in vivo. The inactivation rates of colchicine-albumin-magnetic particles and free colchicine in vitro were 56.16% And 58.46%. The antitumor rates of EMT_6 breast cancer xenografts in nude mice were 32.8% and 23.6%, respectively, suggesting that colchicine-magnetic particles plus magnetic field guidance could better inhibit breast cancer in nude mice.
