从水曲柳中克隆一个WRKY基因,序列比对后命名为Fm WRKY40,基因编码区全长为936 bp,编码331个氨基酸(GenBank登录号:KU928310);含有一个WRKY保守结构域和一个C2H2锌指结构域,属于WRKY家族第Ⅱ类成员;分子进化分析表明,Fm WRKY40与芝麻WRKY40亲缘关系最近,蛋白相似性为73%;qRT-PCR表达模式分析表明:Fm WRKY40在干旱胁迫和ABA(脱落酸)胁迫下明显上调表达,该基因在水曲柳应答干旱和其他非生物胁迫中可能起重要的调控作用。
A WRKY gene was cloned from Fraxinus mandshurica. The sequence of the WRKY gene was named Fm WRKY40. The full length of the gene was 936 bp encoding 331 amino acids (GenBank accession number: KU928310), containing a WRKY conserved domain and a C2H2 zinc Finger domain, belonging to the second member of the WRKY family. Phylogenetic analysis showed that Fm WRKY40 had the closest genetic relationship with sesame WRKY40, and the protein similarity was 73%. Analysis of qRT-PCR expression pattern showed that Fm WRKY40 had a close relationship between drought stress and ABA Acid), which may play an important regulatory role in the response to drought and other abiotic stresses in Ash.
