目的:采用反相离子对高效液相色谱结合波长切换技术,测定复方苦参水杨酸散中苦参碱,氧化苦参碱,水杨酸,苯甲酸等4个成分的含量。方法:采用Agilent ZORBAX SB-C18色谱柱(150 mm×4.6 mm,5μm),以乙腈-0.1%磷酸溶液(16∶84)(每100 ml中加庚烷磺酸钠0.2 g)为流动相,流速:1.0 ml·min~(-1);检测波长:0~12 min,220 nm;12~25 min,280 nm;柱温30℃;进样量10μl。结果:苦参碱,氧化苦参碱,水杨酸,苯甲酸的线性范围分别是0.006 030~0.120 6μg(r=0.999 4),0.016 56~0.331 2μg(r=0.999 9),0.717 1~14.34μg(r=0.999 9),0.512 0~10.24μg(r=0.999 9);平均加样回收率分别为98.14%,97.20%,97.05%,98.39%,RSD分别为1.38%,0.32%,0.81%,1.26%(n=6)。结论:该方法快速,准确,可用于复方苦参水杨酸散中苦参碱,氧化苦参碱,水杨酸,苯甲酸等4个成分的含量测定。
OBJECTIVE: To determine the contents of four components of matrine, oxymatrine, salicylic acid and benzoic acid in Compound Kushen Salicylic Acid Powder by reversed-phase ion-pair high performance liquid chromatography combined with wavelength-switching technique. METHODS: The mobile phase consisted of Agilent ZORBAX SB-C18 column (150 mm × 4.6 mm, 5 μm) with acetonitrile-0.1% phosphoric acid solution (16:84) (0.2 g heptane sulfonate per 100 ml) Flow rate: 1.0 ml · min -1; detection wavelength: 0-12 min, 220 nm; 12-25 min, 280 nm; column temperature 30 ℃; injection volume 10μl. Results: The linear ranges of matrine, oxymatrine, salicylic acid and benzoic acid were 0.006 030 ~ 0.120 6μg (r = 0.999 4), 0.016 56 ~ 0.331 2μg (r = 0.999 9), 0.717 1 ~ 14.34 The average recoveries were 98.14%, 97.20%, 97.05% and 98.39%, respectively, with RSDs of 1.38%, 0.32% and 0.81%, respectively , 1.26% (n = 6). Conclusion: The method is rapid and accurate, and can be used for the determination of four components of matrine, oxymatrine, salicylic acid and benzoic acid in Compound Sophora flavescens Salivarius.
