目的:观察香蕉皮多糖对体外培养的人乳腺癌细胞增殖、克隆形成及其核因子-κB(nudear-kappa B,NF-κB)p65蛋白表达的影响。方法:采用溴化四甲基偶氮[3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide,MTT]法,取对数生长期人乳腺癌细胞(MCF-7),随机分为药物组与对照组,用质量浓度为0.5,1.5,2.5,3.5 g·L-1香蕉皮多糖作用于MCF-7细胞,分别干预24,48,72 h。通过克隆形成试验,Western blot试验,观察香蕉皮多糖对MCF-7细胞克隆形成及对MCF-7细胞NF-κB(p65)蛋白表达的影响,试验中设置药物组与对照组,药物组MCF-7细胞加入质量浓度为0.5,1.5,2.5 g·L-1香蕉皮多糖干预48 h。结果:香蕉皮多糖对MCF-7细胞的增殖有抑制作用,诱导肿瘤细胞凋亡作用与药物浓度和药物作用时间成正相关。MCF-7细胞的克隆形成率,NF-κB(p65)蛋白的表达随香蕉皮多糖浓度升高而显著下降。结论:香蕉皮多糖对MCF-7细胞增殖,克隆形成有抑制作用,(P<0.05)。香蕉皮多糖干预后,MCF-7细胞表达NF-κB(p65)明显减弱。其诱导MCF-7细胞凋亡作用机制可能下调NF-κB表达水平有关。
Objective: To observe the effects of banana peel polysaccharides on the proliferation, colony formation and the expression of p65 protein of nudear-kappa B (NF-κB) in cultured human breast cancer cells in vitro. Methods: Human breast cancer cells (MCF-7) were cultured in logarithmic growth phase using MTT assay. , Were randomly divided into drug group and control group. MCF-7 cells were treated with 0.5, 1.5, 2.5 and 3.5 g · L-1 banana peel polysaccharides, respectively, for 24, 48 and 72 h. The formation of MCF-7 cells and the expression of NF-κB (p65) protein in MCF-7 cells were observed by clonogenic assay and Western blot assay. The drug group and the control group were set up. MCF- 7 cells were treated with 0.5,1.5,2.5 g · L-1 banana peel polysaccharides for 48 h. Results: Banana peel polysaccharides inhibited the proliferation of MCF-7 cells and induced the apoptosis of tumor cells in a dose-dependent manner. The colony formation rate of MCF-7 cells and the expression of NF-κB (p65) protein decreased significantly with the increase of the concentration of banana peel polysaccharides. CONCLUSION: Banana peel polysaccharide can inhibit the proliferation and colony formation of MCF-7 cells (P <0.05). After the intervention of banana peel polysaccharide, the expression of NF-κB (p65) in MCF-7 cells was obviously weakened. The mechanism of its induction of apoptosis in MCF-7 cells may be related to the down-regulation of NF-κB expression.
