目的探讨脐血细胞输注对急性心肌梗死(AMI)大鼠的治疗作用,为脐血应用于心血管疾病的临床治疗提供实验依据。方法将30只Wistar大鼠随机分为正常组、模型组和治疗组(10只/组),模型组和治疗组腹腔注射异丙肾上腺素4mg/kg.d,连续2 d,复制大鼠AMI模型。将脐血细胞体外用DAPI标记后,经尾静脉输入治疗组大鼠体内,模型组给予等量无血清培养基,3周后超声心动图检测大鼠心功能情况,心脏标本连续冰冻切片观察标记的脐血细胞,石蜡切片HE染色观察心脏病理改变。结果AMI大鼠输注脐血细胞3周后,治疗组心肌组织冰冻切片可观察到DAPI荧光标记阳性细胞存在;与模型组比较,治疗组心功能明显改善(P<0.05),HE染色可见心肌充血及炎症减轻,心肌细胞萎缩和心肌瘢痕组织明显减少。结论在未使用免疫抑制剂的情况下,脐血细胞经尾静脉输注治疗后可归巢至AMI大鼠心脏,并能改善心功能,脐血治疗有望成为治疗AMI的有效手段。
Objective To investigate the therapeutic effect of umbilical cord blood transfusion on acute myocardial infarction (AMI) in rats and provide experimental evidence for clinical application of cord blood in the treatment of cardiovascular diseases. Methods Thirty Wistar rats were randomly divided into normal group, model group and treatment group (10 rats / group). Rats in model group and treatment group were injected intraperitoneally with isoproterenol 4mg / kg.d for 2 days, model. The umbilical cord blood cells were labeled with DAPI in vitro and the rats in the untreated group were given the same amount of serum-free medium through the tail vein. After three weeks, the cardiac function of the rats was examined by echocardiography. Umbilical cord blood cells, paraffin sections HE staining observed pathological changes. Results Three weeks after infusion of cord blood cells in AMI rats, DAPI positive cells were observed in the frozen sections of myocardial tissue in the treatment group. Compared with the model group, the cardiac function of the treated group was significantly improved (P <0.05) And reduce inflammation, myocardial cell shrinkage and myocardial scar tissue was significantly reduced. Conclusion In the absence of immunosuppressive agents, umbilical cord blood cells can be homed to the heart of AMI rats after transfusion through the tail vein, and can improve cardiac function. Cord blood therapy is expected to be an effective treatment for AMI.
