目的 探索结核菌素纯蛋白衍生物 (purifiedproteinderivative ,PPD)对卵蛋白 (ovalbumin ,OVA)致敏小鼠肺组织TH1 细胞的在体调节和对气道炎症的作用。方法 取 37只Balb c小鼠 ,雌雄兼有 ,6~ 8周。OVA雾化连续 1 0d ,每次雾化2 0min ,复制OVA致敏模型。观察记录小鼠耗氧量、肺组织淋巴细胞反应和支气管肺泡灌洗液 (bronchoalveolarlavagefluid,BALF)中炎性细胞变化。结果 OVA致敏组小鼠肺组织CD4+T淋巴细胞增加 ,CD8+T淋巴细胞无明显变化 ,主要表现为γ IFN阴性CD4+T淋巴细胞的增加 ,γ IFN+ CD4比例降低。PPD治疗后 ,肺组织CD4+淋巴细胞降低 ,但γ IFN+细胞明显增加。OVA致敏小鼠消耗氧增加 ,PPD治疗后降低。结论 使用本实验体系PPD可上调TH1 细胞并减轻实验性哮喘的气道反应
Objective To explore the effect of purified protein derivative (PPD) on the regulation of airway inflammation and airway inflammation in lung tissue of mice sensitized with ovalbumin (OVA). Methods Taken 37 Balb c mice, both male and female, for 6 to 8 weeks. OVA aerosol continuous 10d, 20min each atomization, replication OVA sensitized model. Oxygen consumption, lung tissue lymphocyte reaction and inflammatory cell changes in bronchoalveolar fluid (BALF) were observed and recorded. Results The CD4 + T lymphocytes in lung tissue of OVA sensitized mice increased and there was no significant change in CD8 + T lymphocytes. The main manifestations were γ IFN negative CD4 + T lymphocytes and γ IFN + CD4 ratio decreased. After PPD treatment, CD4 + lymphocytes in lung tissue decreased, but γ IFN + cells increased significantly. OVA sensitized mice increased oxygen consumption, reduced after PPD treatment. Conclusion PPD can up-regulate TH1 cells and reduce airway response in experimental asthma
