目的研究六月青石油醚萃取物(petroleum ether extract of LYQ,LYQPE)对人胃腺癌SGC-7901细胞凋亡的影响及其部分作用机制。方法用不同浓度的LYQPE作用于SGC-7901,作用48h后,流式细胞术检测对细胞凋亡及其凋亡相关蛋白p53影响;RT-PCR检测凋亡相关基因p53mRNA和黏附基因CD44mRNA的变化;电镜观察超微结构变化。结果 LYQPE体外作用于SGC-7901,在40~320μg/ml时呈不同程度的促进凋亡;呈剂量依赖性的抑制p53蛋白、mRNA水平和CD44mRNA水平的表达;在160μg/ml时,电镜观察可见典型的凋亡形态学改变。结论 LYQPE对SGC-7901有明显促凋亡作用;LYQPE抑制SGC-7901p53蛋白、mRNA水平和CD44mRNA水平的表达有可能是促进SGC-7901凋亡的作用机制。
Objective To study the effect of ester ether extract of LYQ (LYQPE) on the apoptosis of human gastric adenocarcinoma SGC-7901 cells and its partial mechanism. METHODS: SGC-7901 cells were treated with different concentration of LYQPE. After 48 hours, flow cytometry was used to detect apoptosis and apoptosis-associated protein p53. RT-PCR was used to detect apoptosis-related gene p53 mRNA and adhesion gene CD44 mRNA. Electron microscopy observed ultrastructural changes. RESULTS: LYQPE acted on SGC-7901 in vitro, and promoted apoptosis to varying degrees at 40-320 μg/ml; it inhibited the expression of p53 protein, mRNA and CD44 mRNA in a dose-dependent manner; at 160 μg/ml, electron microscopy showed that Typical morphological changes in apoptosis. Conclusions LYQPE has obvious pro-apoptotic effect on SGC-7901. The inhibition of SGC-7901p53 protein, mRNA and CD44 mRNA expression by LYQPE may be the mechanism that promotes the apoptosis of SGC-7901.
