目的 对河南省汉族群体的 8个短串联重复序列 (short tandem repeats,STR)基因座等位基因频率进行研究 ,得到河南群体 TH0 1、FES、D19S40 0、D7S82 0、D16 S5 39、D2 0 S16 1、D3S15 45和 D5 S818基因座的群体遗传学数据。方法 EDTA抗凝血样采自河南 117名无血缘关系汉族个体 ,酚氯仿法抽提DNA,PCR扩增 ,非变性聚丙烯酰胺垂直凝胶电泳 ,银染显色分析。结果 得到 8个基因座的等位基因频率 ,各基因座的杂合度分别为 :0 .6 6、0 .6 7、0 .80、0 .76、0 .79、0 .79、0 .78、0 .78;个体识别率 :0 .83、0 .83、0 .94、0 .91、0 .93、0 .93、0 .92、0 .92。结论 8个 STR基因座具有较高的杂合度 ,等位基因分布符合 Hardy-Weinberg平衡 ,是较理想的遗传标记 ,可用于法医学个体识别和亲权鉴定
Objective To study the allele frequencies of 8 short tandem repeats (STR) loci in Han population in Henan Province, and to obtain the frequencies of TH0 1, FES, D19S40 0, D7S82 0, D16 S5 39, D2 0 S16 1, D3S15 45 and D5 S818 loci. Methods EDTA anticoagulant blood samples were collected from 117 unrelated Han individuals in Henan Province. DNA was extracted by phenol - chloroform method, and PCR amplification was carried out. The polyacrylamide gel electrophoresis was performed by vertical gel electrophoresis and the silver staining was performed. Results The allele frequencies of 8 loci were obtained, and the heterozygosity of each locus was: 0.666.0, 6.77, 0.80, 0.76, 0.79, 0.79, 0.78 , 0 .78; individual recognition rate: 0 .83,0 .83,0 .94,0 .91,0 .93,0 .93,0 .92,0 .92. Conclusion The eight STR loci are highly heterozygous, allele distribution is in line with Hardy-Weinberg equilibrium, which is an ideal genetic marker for forensic identification and paternity testing
