目的:从自然衰老大鼠海马神经元凋亡角度探索补肾方的作用机制。方法:以自然衰老SD雄性大鼠(24月龄)为肾虚衰老模型,大鼠随机分老年组(模型)、左归组和右归组,另设青年组,共四组。大鼠自20月龄始,左归组和右归组分别给予左、右归丸,均按相当于生药7.5g/kg(成人临床用量的15倍)自由饮用稀释药液,每周停药2天,连续4个月;原位杂交检测海马区神经元Bax、Bcl-2 mRNA表达。结果:与青年组比较,老年组大鼠海马CA1区Bcl-2基因mRNA表达水平显著下降(P<0.05),Bax基因mRNA表达水平显著升高(P<0.05);与老年组比较,左归组和右归组大鼠海马CA1区Bax、Bcl-2基因mRNA表达水平均有显著变化(P<0.05)。结论:自然衰老大鼠的海马CA1区神经细胞凋亡相关基因表达异常,补肾方可通过调节海马CA1区神经细胞Bax、Bcl-2基因mRNA的表达,降低海马神经细胞凋亡,进而改善海马功能、延缓衰老。
Objective: To explore the mechanism of Bushen Recipe from the perspective of apoptosis of hippocampal neurons in natural aging rats. Methods: Male SD rats aged 24 months were randomly divided into four groups. The rats were randomly divided into the aged group (model), Zuogui group and Yougui group, and the other four groups. Rats from the age of 20 months, Zuogui group and Yougui group were left and right for the pill, are equivalent to 7.5g / kg of crude drug (15 times the clinical dose of adult) free to drink diluted solution, weekly withdrawal 2 days for 4 months. The expressions of Bax and Bcl-2 mRNA in hippocampal neurons were detected by in situ hybridization. Results: Compared with the young group, the expression of Bcl-2 mRNA in hippocampal CA1 region was significantly decreased (P <0.05) and the mRNA expression of Bax was significantly increased in the aged group (P <0.05). Compared with the aged group, The mRNA expression of Bax and Bcl-2 in hippocampal CA1 region of rats in both groups were significantly changed (P <0.05). Conclusion: The expression of neuronal apoptosis-related genes in hippocampus CA1 of naturally aged rats is abnormal. Bushen can reduce the apoptosis of hippocampus neurons by regulating the expression of Bax and Bcl-2 mRNA in hippocampal CA1 area, and then improve the function of hippocampus ,anti aging.
