黄芪桂枝五物汤中总皂苷对小鼠部分免疫功能的影响

目的探讨黄芪桂枝五物汤中总皂苷对机体免疫功能的影响,从而为相关产品开发和临床应用提供实验依据。方法取40只小鼠,随机分为皂苷组1(低剂量),皂苷组2(中等剂量)和皂苷组3(高剂量)以及正常对照组,每组10只。总皂苷组每天灌服0.2ml浓度为5,10,20mg/ml的总皂苷溶液,正常对照组每天灌服0.2ml生理盐水,10d后处死,取样测定小鼠各项免疫指标。1检测小鼠巨噬细胞吞噬功能;2小鼠后足迟发型超敏反应强度;3血凝法测定小鼠血清中溶血素含量;4对脾脏和胸腺称重计算脾脏指数和胸腺指数。结果皂苷各组小鼠巨噬细胞吞噬率和吞噬指数明显低于对照组(P<0.01);皂苷各组小鼠左右后足重量差与对照组小鼠相比,有明显降低(P<0.01);皂苷各组小鼠抗体积数低于对照组(P<0.05);皂苷各组小鼠脾脏指数和胸腺指数均较对照组小鼠有明显降低(P<0.01)。结论黄芪桂枝五物汤中总皂苷可以显著降低机体的部分固有免疫功能、细胞免疫功能和体液免疫功能。 Objective To investigate the effects of total saponins from Astragalus and Guizhi Wuwu Decoction on the immune function and to provide experimental basis for the development of related products and clinical application. Methods Forty mice were randomly divided into four groups: saponin group 1 (low dose), saponin group 2 (medium dose) and saponin group 3 (high dose), and normal control group, 10 mice in each group. The total saponin group was given 0.2 ml of total saponin solution with the concentration of 5, 10 and 20 mg / ml every day. The normal control group was given 0.2 ml normal saline daily and sacrificed after 10 days. The immune indexes of the mice were determined by sampling. 1 detection of macrophage phagocytosis in mice; 2 hindpaw delayed hypersensitivity; 3 hemagglutination assay of hemolysin in mouse serum; 4 spleen and thymus were weighed to calculate the spleen index and thymus index. Results The phagocytic rate and phagocytic index of macrophages of mice in each group were significantly lower than those in the control group (P <0.01). The weight differences of the left and right hind paws in the saponin groups were significantly lower than those in the control group (P <0.01) (P <0.05). The spleen index and thymus index of mice in each saponin group were significantly lower than those in the control group (P <0.01). Conclusion Total saponin of Huangqi Guizhi Wuwu Decoction can significantly reduce the part of the body’s innate immune function, cellular immune function and humoral immune function.
