目的观察牛磺酸对过度训练大鼠心肌细胞凋亡及fas和caspase-3基因表达的影响,探讨牛磺酸对运动心脏保护作用的可能机制。方法采用力竭性游泳训练建立大鼠过度训练模型,30只SD大鼠随机等分为正常对照组、过度训练组和过度训练+牛磺酸组。实验后采用流式细胞仪检测各组大鼠心肌细胞凋亡,半定量RT-PCR检测caspase-3和fas mRNA表达,二步法免疫组化检测Fas和Caspase-3蛋白表达。结果过度训练组大鼠心肌细胞凋亡率、fas和caspase-3基因表达水平均显著高于正常对照组(P<0.01)。而过度训练+牛磺酸组心肌细胞凋亡率、fas和cas-pase-3基因表达与正常对照组无显著差异(P>0.05)。结论牛磺酸对过度训练大鼠心肌细胞凋亡以及fas和caspase-3基因表达增高有较好的颉颃作用。
Objective To observe the effect of taurine on cardiomyocyte apoptosis and the expression of fas and caspase-3 in over-trained rats, and to explore the possible mechanism of taurine on exercise-induced cardioprotection. Methods Excessive swimming training was used to establish an over-training model in rats. Thirty SD rats were randomly divided into normal control group, over-training group and over-training + taurine group. After the experiment, the apoptosis of cardiomyocytes in each group was detected by flow cytometry. The expression of caspase-3 and fas mRNA was detected by semi-quantitative RT-PCR. The expression of Fas and Caspase-3 protein was detected by two-step immunohistochemistry. Results The apoptosis rate of cardiomyocytes, fas and the expression of caspase-3 gene in over-training group were significantly higher than those in normal control group (P <0.01). However, there were no significant differences in Fas and caspase-3 gene expression between the over-training + taurine group and normal control group (P> 0.05). Conclusion Taurine has a good antagonistic effect on myocardial cell apoptosis and fas and caspase-3 gene expression in over-trained rats.
