目的探讨新型小分子Nutlins结构类似物NL32在体外对非小细胞肺癌A549细胞增殖及凋亡的影响。方法采用MTT法检测不同浓度NL32(100、33、11、3.7、1.2、0.4、0.14μmol/L)对A549细胞增殖的影响,并计算其半数致死浓度(IC50);流式细胞术检测NL32对A549细胞凋亡及细胞周期的影响;Western blot法检测NL32对A549细胞p53蛋白及其下游蛋白p21表达的影响。结果不同浓度的NL32对A549细胞的增殖均有一定的抑制作用,且呈剂量依赖性,其对A549细胞的IC50为15.9μmol/L;NL32可诱导A549细胞凋亡,并将细胞周期阻滞在G2期,且呈剂量依赖性;NL32能上调A549细胞p53和p21蛋白的表达。结论 NL32能有效地以浓度依赖的方式抑制A549细胞增殖,诱导细胞凋亡,引起细胞发生G2期周期阻滞,NL32上调p53和p21蛋白的表达可能是其引发A549细胞周期阻滞和凋亡的原因之一。
Objective To investigate the effect of a novel small molecule Nutlins structural analog NL32 on proliferation and apoptosis of non-small cell lung cancer A549 cells in vitro. Methods MTT assay was used to determine the effect of NL32 (100,33,11,3.7,1.2,0.4,0.14μmol / L) on the proliferation of A549 cells and the median lethal concentration (IC50) of the cells was calculated. Flow cytometry A549 cell apoptosis and cell cycle. Western blot was used to detect the effect of NL32 on p53 protein and its downstream protein p21 expression in A549 cells. Results Different concentrations of NL32 inhibited the proliferation of A549 cells in a dose-dependent manner. The IC50 of A549 cells was 15.9μmol / L. NL32 induced the apoptosis of A549 cells and arrested cell cycle in G2 phase in a dose-dependent manner. NL32 up-regulated p53 and p21 protein expression in A549 cells. Conclusion NL32 can effectively inhibit proliferation and induce apoptosis of A549 cells in a concentration-dependent manner, resulting in cell cycle arrest of G2 phase. NL32 up-regulates the expression of p53 and p21 proteins possibly by inducing cell cycle arrest and apoptosis in A549 cells one of the reasons.
