目的观察高血压大鼠脑缺血再灌注后胰岛素样生长因子(IGF)-Ⅰ、B细胞淋巴瘤(Bcl)-2表达与细胞凋亡的关系。方法制备高血压大鼠脑缺血再灌注模型,用免疫组织化学染色法及流式细胞术分别观察脑组织中IGF-Ⅰ、Bcl-2的表达情况及细胞凋亡。结果同对照组相比缺血再灌注组神经细胞凋亡率增高(P<0.01);IGF-Ⅰ与Bcl-2的表达均升高(P<0.01),IGF-Ⅰ比Bcl-2升高显著(P<0.05)。结论IGF-Ⅰ、Bcl-2同时参与了脑缺血再灌注后的细胞凋亡;IGF-Ⅰ可能通过调控Bcl-2发挥抗凋亡作用。
Objective To observe the relationship between insulin-like growth factor (IGF-Ⅰ), Bcl-2 expression and apoptosis in spontaneously hypertensive rats after cerebral ischemia-reperfusion. Methods The model of cerebral ischemia-reperfusion in hypertensive rats was made. The expression of IGF-Ⅰ and Bcl-2 in brain tissue and the apoptosis were observed by immunohistochemical staining and flow cytometry respectively. Results Compared with the control group, the apoptotic rates of neuronal cells in ischemia-reperfusion group were significantly increased (P <0.01), the expressions of IGF-Ⅰ and Bcl-2 were increased (P <0.01) Significant (P <0.05). Conclusions Both IGF-Ⅰ and Bcl-2 participate in apoptosis after cerebral ischemia-reperfusion. IGF-Ⅰ may play an anti-apoptotic role by regulating Bcl-2.
