目的:对大鼠原位肠肝灌流模型的灌流液组成成分进行优化,以获取最佳灌流液配方。方法:根据灌流液配方不同,共分8组。观察各组灌流液的pH、乳酸脱氢酶(LDH)、丙氨酸氨基转移酶(ALT)活性,以及肝、肠干湿重比,从而比较不同灌流液对肝功能、肝细胞损伤以及组织水肿程度的影响。结果:第8组灌流液(不加红细胞、地塞米松、去甲肾上腺素组)的pH,LDH,ALT与第1组灌流液(加红细胞、地塞米松、去甲肾上腺素组)相比无显著性差异;第8组灌流液肠干湿重比与正常对照组相比无显著性差异;结论:红细胞、地塞米松、去甲肾上腺素对于维持肠肝灌流模型功能无显著影响。
OBJECTIVE: To optimize the composition of perfusate in rat intestinal perfused liver in order to obtain the best perfusate formula. Methods: According to different perfusion solution, divided into 8 groups. The pH, lactate dehydrogenase (LDH), alanine aminotransferase (ALT) activity, as well as the ratio of wet to dry weight of liver and intestine in each group were observed to compare the effects of different perfusate on hepatic function, hepatocellular injury and tissue Edema effect. Results: The pH, LDH, ALT in the eighth group of perfusate (without red blood cells, dexamethasone, norepinephrine group) compared with the first group perfusate (plus red blood cells, dexamethasone, norepinephrine group) There was no significant difference between the two groups. There was no significant difference in the intestinal wet weight ratio between the 8th group and the control group. Conclusion: Erythrocyte, dexamethasone and norepinephrine had no significant effect on the intestinal perfusion model.
