目的研究急性肺栓塞时结合珠蛋白和铁蛋白的表达变化。方法采用RT-PCR方法检测急性肺栓塞大鼠模型肺组织中结合珠蛋白和铁蛋白mRNA水平的变化;采用Westernblot技术检测急性肺栓塞大鼠血清中结合珠蛋白和铁蛋白蛋白水平的变化;采用免疫散射比浊法测定58位肺栓塞/深静脉血栓(PE/DVT)患者和40例对照(对照组)血清中的结合珠蛋白和铁蛋白的蛋白水平。结果急性肺栓塞后大鼠肺组织中结合珠蛋白和铁蛋白mRNA水平逐渐升高,血清中结合珠蛋白和铁蛋白的蛋白水平也逐渐升高。PE/DVT病人血清中结合珠蛋白的蛋白浓度为2063·48±425·38mg/L,对照组为824·37±235·24mg/L(P<0·01);PE/DVT患者血清中铁蛋白的蛋白浓度为554·43±136·18μg/L,对照组为79·42±31·57μg/L(P<0·01)。结论急性肺栓塞后肺组织细胞增加了结合珠蛋白和铁蛋白的合成,导致这两种蛋白的血清水平明显升高,表明结合珠蛋白和铁蛋白对PE/DVT病人具有一定的诊断价值。
Objective To investigate the expression of haptoglobin and ferritin in acute pulmonary embolism. Methods The mRNA expression of haptoglobin and ferritin in the lung tissue of acute pulmonary embolism rat model was detected by RT-PCR. The changes of haptoglobin and ferritin protein in the serum of rats with acute pulmonary embolism were detected by Western blot. Immunoblot turbidimetry was used to determine the serum levels of haptoglobin and ferritin in 58 PE / DVT patients and 40 controls (control group). Results After acute pulmonary embolism, the levels of haptoglobin and ferritin mRNA in lung tissue of rats were gradually increased, and the levels of serum haptoglobin and ferritin protein were also increased. The serum concentration of haptoglobin in patients with PE / DVT was 2063.48 ± 425.38mg / L and the control group was 824.37 ± 235.24mg / L (P <0.01). The levels of serum ferritin The protein concentration was 554.43 ± 136.18μg / L and the control group was 79.42 ± 31.57μg / L (P <0.01). Conclusions The lung tissue cells after acute pulmonary embolism increase the synthesis of haptoglobin and ferritin, resulting in significantly increased serum levels of these two proteins, indicating that haptoglobin and ferritin have some diagnostic value in patients with PE / DVT.
