树状大分子对肿瘤有显著的EPR效应,本文在5代聚酰胺-胺类树状大分子表面连接对肿瘤有主动靶向作用的抗生物素蛋白(亲和素),然后用放射性同位素99mTc标记这种兼具被动靶向与主动靶向作用的纳米粒子。Hela细胞体外摄取实验表明连接靶向分子亲和素(Avidin)的Biotin-PAMAM G5-1B4M-99mTc纳米复合物对Hela细胞的摄取远高于未连接靶向分子Avidin的同类纳米粒子。如进一步在小鼠SPECT成像试验中表现出良好的显像效果,这种同位素标记的纳米粒子将有望用于肿瘤的早期诊断或治疗。
The dendrimer has significant EPR effect on the tumor. In this study, avidin (avidin) with active targeting to tumor was attached on the surface of fifth-generation polyamidoamine dendrimer, and then radioisotope 99mTc Labeling this kind of nanoparticles with both passive and active targeting. In vitro uptake experiments of Hela cells showed that uptake of Hela cells by Biotin-PAMAM G5-1B4M-99mTc nanocomplexes, which target the avidin, was much higher than that of the non-targeted Avidin-like nanoparticles. As further demonstrating good imaging in mouse SPECT imaging experiments, this isotope-labeled nanoparticle is expected to be used for the early diagnosis or treatment of tumors.
