目的:探讨葛根芩连汤不同配伍对其主要化学成分甘草酸含量的变化的影响差异。方法:采用反相高效液相色谱法(RP-HPLC)对葛根芩连汤不同配伍前后其主要有效成分甘草酸含量进行测定,测定波长为250nm,色谱柱为Zorbax SB-C18(4.6mm×250mm,5μm),流动相为甲醇:醋酸铵:冰醋酸(67:33:1),柱温20℃,流速为0.7ml/min,进样量为10μl。结果:对照品溶液测定过程中,在RP-HPLC条件下线性关系良好、精密度试验、稳定性试验、重现性试验、回收试验均显示良好的情况下对不同配伍的样品溶液中甘草酸含量进行测定,结果显示甘草酸含量由高到低排序为:甘草(0.89)>甘草、黄芩和葛根(0.59)>甘草和黄芩(0.48)>甘草、葛根和黄连(0.25)>甘草、黄芩、黄连和葛根(0.25)。结论:单味甘草所含甘草酸含量最高,与其他药味配伍可显著降低甘草酸含量,其中方剂配伍中无黄连配伍对甘草酸含量影响较小,临床中有必要全面科学掌握中药方剂的配伍规律,为临床合理用药提供理论依据。
Objective: To investigate the effect of different combinations of Gegen Qinlian Decoction on glycyrrhizic acid content changes of its main chemical components. Methods: The content of glycyrrhizic acid was determined by reversed-phase high-performance liquid chromatography (RP-HPLC) before and after the different compatibility of Gegen Qinlian Decoction. The wavelength was 250nm and the column was Zorbax SB-C18 (4.6mm × 250mm , 5μm). The mobile phase consisted of methanol: ammonium acetate: glacial acetic acid (67: 33: 1) and the column temperature was 20 ℃. The flow rate was 0.7ml / min. Results: During the determination of reference solution, the linear relationship was good under the conditions of RP-HPLC. The results of precision test, stability test, reproducibility test and recovery test showed that the content of glycyrrhizic acid The results showed that the contents of glycyrrhizic acid from high to low were: licorice, skullcap and kudzu (0.59)> licorice and skullcap (0.48)> licorice, kudzu root and coptis (0.25)> licorice root, And Pueraria (0.25). Conclusion: The content of glycyrrhizic acid contained in single licorice is the highest, compatibility with other medicinal herbs can significantly reduce the content of glycyrrhizic acid. The compatibility of herbs without Coptidis Rhizoma has little effect on the content of glycyrrhizin. It is necessary to grasp the compatibility of traditional Chinese medicine , To provide a theoretical basis for clinical rational use of drugs.
