目的为加强流感病毒H7N9灭活疫苗的免疫效果,本研究以BALB/c小鼠为模型,比较了Al(OH)3、poly(I∶C)、CpG、PEI四种佐剂对H7N9全病毒灭活疫苗和裂解疫苗的免疫增强效果。方法将四种佐剂分别与同剂量的流感病毒H7N9全病毒灭活疫苗或裂解疫苗联合免疫,在肌内注射免疫3周后使用40×LD50的同源病毒攻击小鼠,通过检测小鼠免疫后的抗体水平及攻毒后的小鼠存活率、体质量丢失情况及肺部病毒滴度来评价疫苗联合佐剂的免疫效果。结果四种佐剂均能提高两种灭活疫苗的抗体水平,此外,裂解疫苗和全病毒疫苗单独免疫时小鼠的存活率分别为33.3%、90.0%。除poly(I∶C)对全病毒苗没有显示出增强保护力的佐剂效果外,其他添加佐剂组小鼠均可达100.0%存活。结论全病毒疫苗+Al(OH)3和裂解疫苗+CpG诱导的免疫效果最佳,表明佐剂效应的发挥,会受到疫苗类型的影响。
In order to enhance the immune effect of H7N9 influenza virus inactivated vaccine, BALB / c mice were used as the model to compare the immunogenicity of H7N9 full-virulent virus with four adjuvants, Al (OH) 3, poly (I: C), CpG and PEI Immune enhancement of inactivated and split vaccines. Methods Four kinds of adjuvants were respectively immunized with the same dose of H7N9 virus inactivated virus or split vaccine and challenged with 40 × LD50 homologous virus 3 weeks after intramuscular injection, After the antibody levels and challenge the survival of mice, the loss of body mass and lung virus titer to evaluate the vaccine vaccine adjuvant immune effect. Results All the four adjuvants increased the antibody levels of the two inactivated vaccines. In addition, the survival rates of the mice immunized with the split vaccine and the whole virus vaccine were 33.3% and 90.0%, respectively. Except for poly (I: C), which did not show any protective effect on the whole virus vaccine, the other adjuvanted mice survived 100.0%. Conclusions The whole virus vaccine + Al (OH) 3 and split vaccine + CpG induced the best immune effect, indicating that the adjuvant effect exerted, will be affected by the type of vaccine.
