细胞滋养层细胞和肿瘤细胞的侵入行为具有惊人的相似性,其中基质金属蛋白酶(MMPs)是滋养层细胞和肿瘤细胞侵入的非常重要的介导因素.近来,MMPs家族的一个新成员── MMP-28被克隆并确定下来,采用反转录-聚合酶链式反应(RT-PCR)、酶谱分析和Northern blot等方法,检测了妊娠早期细胞滋养层细胞和绒毛膜癌细胞系JEG-3细胞中MMP-28的基因表达和蛋白分泌情况.RT-PCR显示,细胞滋养层细胞和JEG-3细胞均有MMP-28 mRNA的表达;Northern blot研究显示,JEG-3细胞中MMP-28 mRNA的表达强于细胞滋养层细胞中MMP-28 mRNA的表达,具有时间依赖关系;酶谱分析显示,JEG-3细胞中MMP-28蛋白分泌活性高于细胞滋养层细胞中MMP-28蛋白分泌活性,呈时间依赖关系,这与MMP-28的基因表达结果相一致.进一步证明了MMP-28可能在正常妊娠和肿瘤发展等一些与组织重建有关的过程中发挥一定的作用.
