实验选用170~210g的雄性Sprague-Dawley大鼠,侧脑室注入秋水仙素80~120μg,48小时后经左心室灌流固定,恒冷箱连续切片,厚5μm,每两个相邻的切片相对应,融表在两组载片上,分别用促肾上腺皮质激素释放因子抗血清(Anti-CRF)和神经降压素抗血清(Anti-NT)孵育,ABC或PAP免疫组织化学反应后,DAB呈色,检查、比较配对切片上免疫反应阳性细胞的位置和形态。结果,在相邻切片上看到卵圆核内有数量不等的位置相同、形态相似的免疫反应阳性细胞,即CRF-NT双标记或两种神经肽共存神经元。本组实验首次发现卵圆核内有CRF-NT共存神经元。
Experimental male Sprague-Dawley rats (170-210g) were injected into the lateral ventricle. Colchicine was injected into the lateral ventricle 80 ~ 120μg. After 48 hours, they were fixed by left ventricular perfusion. The continuous cold box was sliced continuously and the thickness was 5μm. This experiment for the first time found in the oval nucleus CRF-NT co-existing neurons.
