以白姜花(Hedychium coronarium)试管苗为外植体,采用薄片培养技术,研究了试管苗苗龄、薄片所在位置、植物生长调节剂的浓度及组成对薄片不定芽诱导的影响。结果表明:从苗龄为28d试管苗的第1条根所在部位切取的薄片,出芽能力最强,每棵苗可获得9.5个芽;在含有3mg/L 6-BA的不定芽诱导培养基中,从1株试管苗最多可获得9.9个不定芽;在含有0.1mg/L TDZ的不定芽诱导培养基中,从1株试管苗最多可获得12.8个不定芽;将所获得的不定芽接种在成苗培养基1/2MS+1g/L活性炭中能有效促进根的发育,植株生长健壮;成熟试管苗驯化后移栽室外,存活率达95%。
In vitro culture of Hedychium coronarium seedlings was used to study the effects of plantlet seedling age, the location of flakes, the concentration of plant growth regulators and the composition on the induction of adventitious buds.
