补肾活血颗粒对维甲酸所致大鼠骨质疏松模型的药效学研究

目的:明确补肾活血颗粒对维甲酸所致大鼠骨质疏松模型的药效作用,并探讨其作用机制。方法:48只SD大鼠随机分为正常组、模型组、补肾活血颗粒低、中、高剂量给药组、仙灵骨葆组6组,除空白组外,其余各组每日ig给予维甲酸70 mg·kg-1,连续14 d。造模与给药同时进行,低、中、高剂量组分别给予补肾活血颗粒流浸膏10,20,40 g·kg-1,仙灵骨葆组给予仙灵骨葆药液0.3 g·kg-1,模型组、正常组ig等容积生理盐水。给药4周后,眼眶取血测血清骨碱性磷酸酶(BALP)、血清骨钙素(BGP)、血清抗酒石酸酸性磷酸酶(TRACP-5β);处死动物后取左、右股骨,左侧用于制作组织病理切片;右侧进行骨密度测定。结果:与正常组相比,模型组大鼠体重增加缓慢(P<0.05),BALP,TRACP-5β显著升高(P<0.01),BGP显著降低(P<0.05),骨密度显著降低(P<0.05)。与模型组比较,各给药组体重增加较快(P<0.05),均可明显下调BALP(P<0.01),补肾活血颗粒流浸膏中、高剂量组及仙灵骨葆组显著提高BGP(P<0.05),降低TRACP-5β(P<0.01),并显著升高骨密度(P<0.05)。结论:补肾活血颗粒具有防治骨质疏松的作用,调节骨生成与骨破坏的紊乱状态,改善骨组织显微结构,提高骨密度和骨更新率。 OBJECTIVE: To determine the pharmacodynamic effects of Bushen Huoxue granule on rat model of osteoporosis caused by retinoic acid and its mechanism of action. Methods: Forty eight Sprague-Dawley rats were randomly divided into six groups: normal group, model group, Bushen Huoxue granule low, medium and high dose administration groups and Xianlinggubao group. Except for the blank group, Formic acid 70 mg · kg-1 for 14 days. At the same time, the rats in low, medium and high dose groups were treated with Bushen Huoxue Granule Flow Extract 10, 20 and 40 g · kg-1, Xianlinggubao Capsule was given Xianling Gubao 0.3 g · kg -1, model group, normal group ig volume of normal saline. Four weeks after the administration, blood orbital bone alkaline phosphatase (BALP), serum BGP and TRACP-5beta were measured. Results: Compared with the normal group, the body weight of model rats increased slowly (P <0.05), BALP and TRACP-5β significantly increased (P <0.01), BGP significantly decreased (P <0.05) <0.05). Compared with the model group, body weight of each administration group increased rapidly (P <0.05), but BALP was significantly decreased (P <0.01). Bushen Huoxue Granule Flow Extract and Xianling Gubao Group significantly increased BGP (P <0.05), decreased TRACP-5β (P <0.01), and significantly increased bone mineral density (P <0.05). Conclusion: Bushen Huoxue Granule can prevent and cure osteoporosis, regulate the disorder of osteogenesis and bone destruction, improve the microstructure of bone tissue and increase the bone density and bone turnover rate.
