目的观察大鼠背根神经节(DRG)中即刻早期基因(IEG s)c-jun在慢性压迫性损伤(CC I)疼痛模型中的变化,探讨慢性神经痛的发生机制。方法将SD大鼠18只,随机分成CC I组和假手术组各9只,CC I组参照B ennett和X ie等方法建立CC I模型,假手术组仅暴露坐骨神经主干,不结扎。于术前2d和术后1、3、5、7d测定两组机械痛阈和热痛阈,术后7d取大鼠双侧L4、L5和L6DRG,免疫组化法分析DRG c-jun变化。结果CC I组损伤侧于术后3d出现热痛觉过敏(P<0.05),术后7d达高峰(P<0.01),且DRG c-jun出现明显表达;而健侧和假手术组没有表达(P<0.01)。结论初级感觉神经元中c-jun表达是神经损伤的标志和始动因素。
Objective To investigate the changes of c-jun immediate junctions (IEGs) c-jun in chronic compression injury (CCI) pain model of rat dorsal root ganglion (DRG) and to explore the mechanism of chronic neuralgia. Methods Eighteen SD rats were randomly divided into CCI group and sham operation group, and each group was given CCI model. The CCI model was established according to B ennett and XeI method. The sham operation group only exposed the sciatic nerve trunk and did not ligate. The mechanical pain thresholds and thermal pain thresholds were measured at 2 days before operation and at 1, 3, 5 and 7 days after operation. L4, L5 and L6 DRG were taken at 7 days after operation, and the changes of DRG c-jun were analyzed by immunohistochemistry. Results Heat hyperalgesia was observed on the injured side of CCI group 3d (P <0.05), and reached the peak on the 7th day after operation (P <0.01), and the expressions of DRG c-jun were significantly higher than those in the contralateral and sham-operated groups P <0.01). Conclusions The expression of c-jun in primary sensory neurons is a marker and initiating factor of nerve injury.
