为观察复方 86 1对HSC -T6细胞基质分解素 1(MMP3 )mRNA表达水平的影响 ,复方 86 1以 0 2 5mg/ml、0 5mg/ml、1 0mg/ml等浓度作用于HSC -T6细胞 48小时 ,以逆转录定量PCR方法测定其对HSC -T6细胞MMP3 mRNA表达水平的影响。结果表明 ,复方 86 10 2 5mg/ml、0 5mg/ml、1 0mg/ml等不同浓度的HSC -T6细胞mRNA表达水平依次为 2 75± 0 35、3 0 0± 0 0 1、3 5 0± 0 71,与空白对照组比较 ,可明显提高MMP3 mRNA表达水平 (P <0 0 5或P <0 0 1)。因此 ,复方 86 1提高HSC -T6细胞MMP3 mRNA表达水平 ,可能是其促进胶原降解 ,抗肝纤维化的作用机理之一。
To observe the effect of compound 86 1 on the expression of MMP3 mRNA in HSC-T6 cells, Compound 86 1 was given to HSC-T6 cells at concentrations of 0 2 5mg / ml, 0 5mg / ml and 10mg / ml After 48 hours, the effect of MMP3 mRNA expression on HSC-T6 cells was determined by reverse transcription polymerase chain reaction (RT-PCR). Therefore, compound 86 1 can increase the expression of MMP3 mRNA in HSC-T6 cells, which may be one of the mechanisms of collagen 861 degradation and anti-hepatic fibrosis.
