以纤维素-三(3,5-二甲苯基氨基甲酸酯)为固定相,系统研究了标题化合物在HPLC系统中的拆分。分别考察了流动相组成、流动相流速、柱温等对拆分效果的影响,并考察了样品的稳定性。建立了以手性OD柱为固定相拆分标题化合物的方法。结果表明:用手性OD柱(25 cm×0.46 cm i.d.),以V(正己烷)∶V(异丙醇)=90∶10为流动相,流速1.0mL/min,柱温20℃,检测波长254 nm时对标题化合物得到了良好的分离,分离度达到1.84。
The resolution of the title compound in HPLC system was studied systematically using cellulose-tris (3,5-dimethylphenylcarbamate) as the stationary phase. The effects of mobile phase composition, mobile phase flow rate and column temperature on the resolution were investigated respectively. The stability of the sample was also investigated. A method for the resolution of the title compound using chiral OD column as the stationary phase was established. The results showed that the mobile phase consisted of chiral OD column (25 cm × 0.46 cm id), V (n-hexane): V (isopropanol) = 90:10, the flow rate was 1.0 mL / min and the column temperature was 20 ℃. The title compound was well separated at a wavelength of 254 nm with a resolution of 1.84.
