目的:用高效液相色谱法测定抗心衰颗粒中黄芪甲苷的含量。方法:以吡啶-苯甲酰氯(2.5:1)为衍生化试剂,采用Supelco ODS C_(18)柱(250mm×4.6 mm,5μm),流动相为甲醇-四氢呋喃-0.2%三乙胺水溶液(90:4:6),流速为1.2 ml·min~(-1),检测波长为230 nm。结果:黄芪甲苷衍生物进样量在0.03~0.93μg范围内与峰面积积分值呈良好线性关系(r=0.9999),平均加样回收率为95.17%,RSD=2.31%(n=6)。结论:该方法简便易行,结果准确,重复性好,可用于测定抗心衰颗粒中黄芪甲苷的含量。
Objective: To determine the content of astragaloside IV in anti-heart failure particles by high performance liquid chromatography. METHODS: A pyridine-benzoyl chloride (2.5:1) was used as the derivatization reagent and a Supelco ODS C 18 column (250 mm×4.6 mm, 5 μm) was used. The mobile phase was methanol-tetrahydrofuran-0.2% aqueous solution of triethylamine (90 : 4:6), the flow rate was 1.2 ml·min -1 and the detection wavelength was 230 nm. Results: Astragaloside IV had a good linear relationship with peak area integral (r=0.9999) in the range of 0.03~0.93μg. The average recovery was 95.17% and RSD was 2.31% (n=6). . Conclusion: The method is simple, accurate and reproducible. It can be used to determine the content of astragaloside IV in anti-heart failure particles.
