目的 从人脑中克隆透明质酸结合蛋白(hbHABP)基因,研究其对各种肿瘤细胞体外增殖的影响。方法 根据已知透明质酸结合蛋白(HABP)DNA序列,设计引物,从人脑cDNA文库中克隆,构建真核细胞表达载体(pcDNA3-hbHABP),转染到人前列腺癌、乳腺癌、肺癌及黑色素瘤细胞中,观察体外细胞生长变化。结果 所有转染pcDNA3-hbHABP的肿瘤细胞相对于转染空载体pcDNA3的对照组细胞,出现生长受抑制现象,在软琼脂上的克隆形成数量也明显减少。结论 hbHABP可能是一种新的抑制肿瘤细胞生长的物质。
Objective To clone the hbHABP gene from human brain and study its effect on the proliferation of various tumor cells in vitro. Methods According to the DNA sequence of known hyaluronan binding protein (HABP), primers were designed and cloned from human brain cDNA library to construct eukaryotic expression vector pcDNA3-hbHABP. The recombinant plasmid was transfected into human prostate cancer, breast cancer, lung cancer and Melanoma cells were observed in vitro changes in cell growth. Results All of the tumor cells transfected with pcDNA3-hbHABP showed inhibition of growth compared with the control cells transfected with empty vector pcDNA3, and the number of clones formed on soft agar was also significantly reduced. Conclusion hbHABP may be a new substance that inhibits the growth of tumor cells.
