【目的】探讨补肾祛风湿中药昆仙胶囊治疗类风湿关节炎(CIA)的作用机理。【方法】选用SPF级Wistar大鼠随机分为正常组,模型组,甲氨蝶呤(MTX)对照组(剂量为0.625 mg.kg-1.周-1)和昆仙胶囊(中药)低、中、高剂量组(剂量分别为0.2、0.4、0.8 g.kg-1.d-1)。采用尾根部皮下多点注射胶原蛋白法复制CIA大鼠模型,测量各组大鼠后肢关节肿胀度,采用酶联免疫吸附(ELISA)法检测大鼠血清中白细胞介素-8(IL-8)的含量,Real-time PCR法检测大鼠滑膜组织IL-8 mRNA的表达水平。【结果】与模型组比较,MTX组、中药各剂量组大鼠滑膜组织IL-8 mRNA表达均显著降低(P<0.05),并可显著降低滑膜组织病理积分(P<0.05或P<0.01);中药高剂量组大鼠血清中IL-8水平显著降低(P<0.05),并可显著降低软骨组织病理积分(P<0.01)。【结论】补肾祛风湿中药昆仙胶囊可能通过抑制大鼠滑膜及血清中IL-8的基因表达,达到抑制大鼠滑膜炎、控制类风湿关节炎炎症活动的目的,且治疗效果与剂量呈正相关。
【Objective】 To investigate the mechanism of the treatment of rheumatoid arthritis (CIA) by tonifying kidney and invigorate the kidney. 【Methods】 SPF Wistar rats were randomly divided into normal group, model group, methotrexate (MTX) control group (0.625 mg.kg-1 wk -1) and Kunxian capsule (traditional Chinese medicine) Medium and high dose group (dose were 0.2,0.4,0.8 g.kg-1.d-1). CIA rats were injected subcutaneously with collagenase at the base of the tail to measure the degree of swelling in the hind limbs of rats in each group. The level of interleukin-8 (IL-8) in rat serum was detected by enzyme-linked immunosorbent assay (ELISA) The content of IL-8 mRNA in synovial tissue of rats was detected by Real-time PCR. 【Results】 Compared with model group, the expression of IL-8 mRNA in synovial tissue of MTX group and traditional Chinese medicine groups were significantly decreased (P <0.05), and significantly decreased the synovial histopathological score (P <0.05 or P < 0.01). The level of IL-8 in the serum of the high-dose Chinese herbal medicine group was significantly decreased (P <0.05), and the pathological score of cartilage tissue was significantly decreased (P <0.01). 【Conclusion】 Bushen Qufengchushi capsule can inhibit the synovitis and the inflammation of rheumatoid arthritis rats by inhibiting the gene expression of IL-8 in synovial membrane and serum of rats, and the therapeutic effect and dose Was positively correlated.
