目的:建立小柴胡汤中黄芩苷、黄芩素、汉黄芩苷、汉黄芩素和甘草酸的HPLC-PDA测定方法。方法:采用反相高效液相色谱法,Symmetry ShieldTMRP18色谱柱(4.6 mm×250 mm,5.0μm);流动相乙腈-0.01%磷酸溶液梯度洗脱;流速0.45 mL.min-1;检测波长251 nm;柱温30℃。结果:本方法的准确度、精密度、重复性、专属性和定量线性范围均符合要求,同时测定了小柴胡汤中黄芩苷、黄芩素、汉黄芩苷、汉黄芩素和甘草酸的含量。结论:该方法简便快速、准确可靠,可用于同时测定小柴胡汤中5种有效成分的含量。
Objective: To establish a HPLC-PDA method for the determination of baicalin, baicalein, baicalin, wogonin and glycyrrhizin in Xiao Chai Hu Tang. Methods: A reversed phase high performance liquid chromatography (HPLC) was used on a Symmetry Shield TMRP18 column (4.6 mm × 250 mm, 5.0 μm). The mobile phase was eluted with a gradient of acetonitrile-0.01% phosphoric acid solution. The flow rate was 0.45 mL · min- ; Column temperature 30 ℃. Results: The accuracy, precision, repeatability, specificity and quantitative linearity of the method met the requirements. The content of baicalin, baicalein, baicalin, wogonin and glycyrrhizin in Xiao Chai Hu Tang was also determined. Conclusion: The method is simple, rapid, accurate and reliable and can be used for the simultaneous determination of five active ingredients in Xiao Chai Hu Tang.
