通过筛选的80种食用植物,对其提取物的抗血栓活性物进行检测,发现丁香花蕾中存在高活性物质,从中提取和纯化出抗血栓多糖类,并对其结构特征进行了分析。 丁香花蕾(1600g)用20L甲醇室温提取5次,过滤后残余物用1.0M氢氧化钠在70℃提取2次,每次2h,得到碱提取物。将碱提取物浸入80%乙醇,用8%十六烷基三甲基溴化铵纯化,用10kDa纤维素柱超滤。渗透液冷冻干燥,残余物再用10kDa纤维素柱超滤,渗透液和残余物分别冷冻干燥制成抗血栓多糖粗品EC-2B和EC-2C。 EC-2B(800mg)通过DEAE-Toyopearl
Through the screening of 80 species of edible plants, the antithrombotic actives of their extracts were detected, and high active substances were found in the buds of cloves. The antithrombotic polysaccharides were extracted and purified from them, and their structural characteristics were analyzed. Clove buds (1600 g) were extracted 5 times with 20 L of methanol at room temperature. After filtering, the residue was extracted twice with 1.0 M sodium hydroxide at 70° C. for 2 h each time to obtain an alkali extract. The base extract was immersed in 80% ethanol, purified with 8% cetyltrimethylammonium bromide, and ultrafiltered with a 10 kDa cellulose column. The permeate was freeze-dried and the residue was ultrafiltered on a 10 kDa cellulose column.
