以葡萄糖 -6 -磷酸脱氢酶 (G6 PD)缺乏红细胞为例 ,研究了氧化、非酶糖基化和高分子聚合物形成并进一步探讨三者之间的可能联系。以高铁血红蛋白、Heinz小体、细胞内还原型谷胱甘肽和膜上巯基含量为指标考察红细胞的氧化程度 ;以间氨基苯硼酸亲和层析法检测红细胞膜蛋白的糖基化率 ;用 SDS聚丙烯酰胺凝胶电泳和凝胶光密度扫描分析检测红细胞膜高分子聚合物的含量。结果显示 ,G6 PD缺乏红细胞发生氧化、非酶糖基化和形成高分子量聚合蛋白 ,且三者之间的可能联系是 :氧化—非酶糖基化—高分子聚合物形成。
Taking glucose - 6 - phosphate dehydrogenase (G6 PD) deficient erythrocytes as an example, the formation of oxidized, non - enzymatic glycation and the formation of polymers were investigated and the possible relationship between them was explored. The degree of erythrocyte oxidation was determined by methemoglobin, Heinz body, intracellular reduced glutathione and the content of sulfhydryl in the membrane. The glycosylation rate of erythrocyte membrane protein was detected by p-aminophenylboronic acid affinity chromatography. SDS polyacrylamide gel electrophoresis and gel optical density scanning analysis of erythrocyte membrane macromolecular polymer content. The results showed that G6-PD lacked erythrocytes oxidative, nonenzymatic glycosylation and formation of high molecular weight aggregates, and the possible linkages among the three were: oxidized-non-enzymatic glycation-polymer formation.
