菌种保存是微生物学中的一项重要工作,其目的在于保存菌种的生活能力,并尽可能维持其原有的生物学特性,为生产、科研和教学及时提供合乎要求的菌种。菌种保存的方法很多,在日常工作中,使用何种方法,应根据微生物本身特性和实验室设备条件而定。归纳起来有两类:一类是将在培养基中生长好的菌种(或加保护剂)移至另外的容器内进行保存,如砂土管保存法,冷冻真空干燥保存法。砂土管保存法适用于细菌的芽胞,霉菌和放线菌的孢子的保存。认为不适宜于保存无芽肥的细菌和酵母菌,而冷冻真空干燥法,由于是在低温真空条件下进行干燥,符合叶天星(1952)氏等归纳的保
The preservation of strains is an important task in microbiology. Its purpose is to preserve the viability of strains and to maintain their original biological characteristics as much as possible so as to provide timely and appropriate strains for production, scientific research and teaching. There are many methods for the preservation of strains. In daily work, which methods are used should be based on the characteristics of the microorganism itself and the conditions of laboratory equipment. To sum up, there are two types: one is to move the bacteria (or protective agent) grown in the culture medium to another container for preservation, such as the sand tube preservation method and freeze vacuum drying preservation method. The sand tube preservation method is suitable for the preservation of spores of bacteria, molds and actinomycetes. It is not considered suitable for the preservation of non-fertilizer-containing bacteria and yeasts, and the freeze-dried vacuum drying method, because it is dried under low-temperature vacuum conditions, meets the needs of induction by Yeh-wing (1952) et al.
