目的探讨人嗅鞘细胞的免疫学特征。方法应用流式细胞仪分析人嗅鞘细胞的相应标记抗原的阳性表达率,用单向混合淋巴细胞反应测定人嗅鞘细胞的组织相容性和免疫耐受性,并与骨髓间充质干细胞对比。结果人嗅鞘细胞与骨髓间充质干细胞表面与移植免疫排斥发生密切相关的标志:HLADR、B71(CD80)、B72(CD86)、CD40和CD40L均为阴性。单向混合淋巴细胞反应显示人嗅鞘细胞、骨髓间充质干细胞均对混合淋巴细胞反应无明显的刺激作用。结论人嗅鞘细胞可能是一种免疫缺陷细胞,在体外有诱导细胞移植免疫耐受性的作用。
Objective To investigate the immunological characteristics of human olfactory ensheathing cells. Methods Flow cytometry was used to analyze the positive expression rates of the corresponding labeled antigen in human olfactory ensheathing cells. The histocompatibility and immunotolerance of human olfactory ensheathing cells were determined by one-way mixed lymphocyte reaction and compared with that of bone marrow mesenchymal stem cells Compared. Results Olfactory ensheathing cells and bone marrow-derived mesenchymal stem cells were closely related to graft rejection. HLADR, B71 (CD80), B72 (CD86), CD40 and CD40L were all negative. Unidirectional mixed lymphocyte reaction showed that human olfactory ensheathing cells and bone marrow mesenchymal stem cells had no obvious stimulating effect on mixed lymphocyte reaction. Conclusion Olfactory ensheathing cells may be an immunocompromised cell, which can induce immune tolerance in vitro.
