将SiO2纳米粒子作为添加剂用于溶菌酶、牛血清白蛋白、牛血红蛋白的毛细管电泳(CE)分离研究。对影响3种蛋白质分离的因素如缓冲溶液的浓度,SiO2纳米粒子、聚合物PVP、乙腈的含量,及分离电压、温度等进行了考察,得到最佳分离条件,即50mmol/L NaH2PO4-10mmol/L H3PO4缓冲溶液(pH=2.8),15%乙腈,10%PVP,0.35%SiO2,分离电压30kV,分离温度25℃,检测波长214nm时,3种蛋白质能得到很好的分离。结果表明,SiO纳米粒子是改善蛋白质毛细管电泳分离的有效添加剂。
Study on Separation of Lysozyme, Bovine Serum Albumin and Bovine Hemoglobin by Capillary Electrophoresis with SiO2 Nanoparticles as Additives. The optimum conditions for the separation of the three kinds of proteins, such as concentration of buffer, concentration of SiO2 nanoparticles, polymer PVP and acetonitrile, separation voltage and temperature, were investigated. The optimum conditions were as follows: 50mmol / L NaH2PO4-10mmol / L H3PO4 buffer solution (pH = 2.8), 15% acetonitrile, 10% PVP and 0.35% SiO2. The separation voltage was 30kV, the separation temperature was 25 ℃ and the detection wavelength was 214nm. The results show that SiO nanoparticles are effective additives to improve the separation of proteins by capillary electrophoresis.
