为了探讨P16基因在人膀胱癌中的肿瘤抑制作用及其在膀胱癌抑癌基因替代治疗中的潜在价值,构建了P16基因表达载体并将其导入有内源性P16基因缺失的人膀肤癌细胞系BIU-87中,观察转染细胞的生长速率、双层软琼脂集落试验及细胞周期的改变,发现转染了P16的细胞生长速率明显降低,软琼脂克隆形成能力显著下降,细胞周期出现向G1/G0期移行的特征性动力学改变。结果表明,P16基因对膀胱癌细胞的恶性增殖具有明显的抑制作用,是膀胱癌抑癌基因替代治疗的一个极有希望的候选靶基因,具有良好的应用前景。
To investigate the role of P16 gene in the suppression of human bladder cancer and its potential value in bladder cancer tumor suppressor gene replacement therapy, a P16 gene expression vector was constructed and introduced into human bladder cancer with loss of endogenous P16 gene. In the cell line BIU-87, the growth rate of the transfected cells, the double-layered soft agar colony test, and the change of the cell cycle were observed. The growth rate of the cells transfected with P16 was significantly reduced, and the ability of cloning of soft agar was significantly decreased, and the cell cycle appeared. Characteristic dynamics of transition to G1/G0 period. The results showed that P16 gene has a significant inhibitory effect on the malignant proliferation of bladder cancer cells, and it is a promising candidate target gene for bladder cancer tumor suppressor gene replacement therapy, and has a good application prospect.
