目的:探讨运动前健脾、运动后补肾对力竭运动大鼠肾细胞凋亡的影响及其分子机制。方法:将24只雄性SD大鼠随机分为安静对照组、力竭运动组、力竭运动+前健脾后补肾组;力竭运动+前健脾后补肾组于力竭运动前40min灌服健脾中药,力竭运动后40min灌服补肾中药,力竭运动组同时灌服生理盐水。安静对照组于安静状态、两运动组在一次性力竭运动后24小时取材,采用透射电镜观察和TUNEL法检测肾凋亡细胞;免疫组化法检测肾细胞凋亡相关蛋白Bcl-2、Bax的表达。结果:力竭运动+前健脾后补肾组与安静对照组比较,肾实质细胞凋亡指数和Bcl-2、Bax蛋白的表达无显著性差异(P>0.05);力竭运动+前健脾后补肾组、安静对照组的肾小管上皮细胞凋亡数、肾细胞凋亡指数和Bax蛋白表达显著低于单纯力竭运动组,差异有统计学意义(P<0.05);三组间的肾细胞Bcl-2蛋白表达无显著性差异(P>0.05)。结论:力竭运动后24小时大鼠肾细胞凋亡显著增加,可能与Bax蛋白表达增加有关;采用运动前健脾、运动后补肾对此具有抑制效果。
Objective: To investigate the effects of invigorating the spleen before and after exercise, and kidney function after exercise on the apoptosis of renal cells in exhaustive exercise rats and its molecular mechanism. Methods: Twenty-four male Sprague-Dawley rats were randomly divided into 4 groups: control group, exhaustive exercise group, exhaustive exercise + Spleen Chinese medicine, exhausted exercise 40min after dredging kidney medicine, exhaustive exercise group while taking saline. Quiet control group in quiet state, the two exercise group in one-time exhaustive exercise 24 hours after drawing, using transmission electron microscopy and TUNEL assay of renal cell apoptosis; immunohistochemical detection of renal cell apoptosis-related proteins Bcl-2, Bax expression. Results: There was no significant difference in apoptotic index and expression of Bcl-2 and Bax between kidney-invigoration group and quiescent control group after exhaustive exercise + before spleen-invigorating group (P> 0.05) The number of renal tubular epithelial cell apoptosis, renal cell apoptosis index and Bax protein expression in post-natal kidney group and quiescent control group were significantly lower than that in purely exhausted exercise group (P <0.05) The expression of Bcl-2 protein had no significant difference (P> 0.05). Conclusion: Apoptosis of rat renal cells increased significantly 24 hours after exhaustive exercise, which may be related to the increase of Bax protein expression. Before exercise, invigorating the spleen and tonifying the kidney can inhibit this effect.
