门冬氨酸钾镁3种注射剂补充钾镁效率和对抗内毒素损伤的比较

目的比较3种不同门冬氨酸钾镁注射剂补充细胞内钾(K+)、镁(Mg2+)的效率及其对抗内毒素(LPS)肝细胞损伤的差异。方法常规培养肝细胞L02株,分为空白对照组、A组、B组和C组。除空白对照组外,各组细胞培养液中分别加入A、B、C三种门冬氨酸钾镁注射剂(均含K+5.3 mmol.L-1及Mg2+2.8 mmol.L-1),继续培养细胞1、2、3、5 h后,测定细胞内K+和Mg2+的含量。对抗内毒素(LPS)损伤实验中将肝细胞L02株分为LPS组、LPS+A组、LPS+B组、LPS+C组,各组均经终浓度为0.1 mg.L-1LPS和20 mmol.L-1的NH4Cl处理,同时将3种门冬氨酸钾镁注射剂分别加入各组,培养2 h后测定细胞培养液中尿素氮浓度和细胞膜Na+-K+-ATP酶的活性。结果与空白对照组比较,各门冬氨酸钾镁注射剂组K+、Mg2+进入细胞内的量均增多,2 h时A组的K+、Mg2+浓度明显高于B组和C组(P<0.05)。在对抗LPS损伤实验中,与LPS组比较,各门冬氨酸钾镁注射剂均可促进氨的清除,表现为尿素生成明显增多,以LPS+A组的效果最为显著(P<0.05);但3种门冬氨酸钾镁对内毒素损伤肝细胞膜Na+-K+-ATP酶活性的效应均不显著(P>0.05)。结论 3种门冬氨酸钾镁注射剂的生物学效应有一定差异,A组能较快地促进K+、Mg2+进入细胞,且显示出较好地拮抗LPS损伤肝细胞的作用。 Objective To compare the efficiency of K + and Mg2 + supplementation of three different potassium magnesium injections and their effects on liver cell injury induced by endotoxin (LPS). Methods L02 hepatocytes were cultured routinely and divided into blank control group, A group, B group and C group. Except for the blank control group, A, B, and C aspartate potassium magnesium injection (containing K + 5.3 mmol.L-1 and Mg2 + 2.8 mmol.L-1) were added into each cell culture medium, After cells were cultured for 1, 2, 3 and 5 h, the contents of K + and Mg2 + in cells were determined. LPS group, LPS + A group, LPS + B group and LPS + C group were divided into LPS group and LPS group with the final concentration of 0.1 mg.L-1 LPS and 20 mmol .L-1 NH4Cl treatment, at the same time, three kinds of magnesium aspartate injection were added into each group. After cultured for 2 h, the concentration of urea nitrogen and the activity of Na + -K + -ATPase in cell membrane were determined. Results Compared with the blank control group, the contents of K + and Mg2 + in the aspartate K + Mg injection group increased, and the concentrations of K + and Mg2 + in group A at 2 h were significantly higher than those in group B and C (P <0.05) . Compared with the LPS group, all potassium magnesium aspartate injection can promote the clearance of ammonia in the anti-LPS injury test, which showed that urea production increased obviously, especially in LPS + A group (P <0.05). The effects of 3 potassium and magnesium aspartate on the activity of Na + -K + -ATPase in liver cell membrane were not significant (P> 0.05). Conclusion The biological effects of three aspartate potassium-magnesium injection are different. A group can quickly promote K +, Mg2 + into the cells, and showed a good antagonism of LPS damage of liver cells.
