作者研究了植物凝集素ConA对红细胞在低粘低切流场中C=0轨道上取向的影响,并采用电子自旋共振(ESR)及荧光偏振技术探讨了其可能的分子基础。实验结果表明:(1)红细胞取向指数OI随ConA(ConcanavalinA)作用浓度呈单调下降趋势,ConA对OI的影响为一可逆现象;(2)ConA作用后的红细胞膜经马来亚酰胺(msl)标记后在ESR谱上表现出强弱固定比(S/W)增高,意味着膜蛋白构象的变化和运动性下降;(3)ConA作用后的红细胞膜脂流动性降低。蛋白构象的改变/膜流动性的下降有可能与红细胞取向能力降低存在着某种内在联系。
The authors studied the effect of the lectin ConA on the orientation of red blood cells in a C = 0 orbit in low viscous low shear flow fields and explored the possible molecular basis by electron spin resonance (ESR) and fluorescence polarization techniques. The results showed that: (1) OI of erythrocyte showed a monotonous decreasing trend with the concentration of ConA (ConcanavalinA), and the effect of ConA on OI was a reversible phenomenon; (2) The membrane of erythrocyte after the action of ConA was treated with maleimide (msl) After labeling, the ESR spectrum shows a strong fixed ratio (S / W), which means the change of membrane protein conformation and the decrease of motility. (3) The membrane fluidity of erythrocytes decreased after ConA treatment. Changes in the conformation of the protein / decrease in membrane fluidity may have some intrinsic relationship to the reduction of erythrocyte orientation.
