CD44与nm23在神经母细胞瘤中的表达及意义

目的检测CD44与nm23基因在神经母细胞瘤(NB)中的表达,探讨CD44和nm23 mRNA的表达与NB发生及转移的关系。方法选择30例术前未经化疗的NB患儿为研究对象,收集术中切除的无坏死新鲜瘤体组织为研究组,相应瘤旁组织为对照组,将标本放置于-80℃冰箱保存。依据Trizol试剂说明书于超净工作台分别提取NB和瘤旁组织mRNA,将其反转录为cDNA后进行扩增内参和引物,凝胶电泳检测nm23 mRNA及CD44 mRNA在NB和瘤旁组织中的表达水平。结果 nm23 mRNA在研究组及对照组中均有表达,但研究组表达低于对照组,研究组中有淋巴结转移组织中的表达低于无淋巴结转移组织中的表达。CD44 mRNA亦在研究组和对照组中均有表达,但研究组表达显著高于对照组,且研究组有淋巴结转移组织中的表达高于在无淋巴结转移组织中的表达。结论 nm23 mRNA及CD44 mRNA的表达水平与NB发生、发展和肿瘤转移密切相关,nm23 mRNA具有抑制NB发生及转移功能,而CD44 mRNA则促进NB的发生及转移。采用PCR技术可以有效检测NB nm23 mRNA及CD44 mRNA的表达水平,有助于判断其生物学行为。 Objective To detect the expression of CD44 and nm23 in neuroblastoma (NB) and to explore the relationship between the expression of CD44 and nm23 mRNA and the occurrence and metastasis of NB. Methods Thirty patients with preoperative NB without chemotherapy were enrolled in this study. Fresh non-necrotic tumor tissue collected during operation was collected as study group and corresponding tumor-adjacent tissue as control group. The specimens were stored in refrigerator at -80 ℃. According to the instructions of Trizol reagent, NB and tumor-adjacent tissue mRNA were respectively extracted from the ultra-clean bench, and then reverse transcribed into cDNA to amplify internal reference and primers. The nm23 mRNA and CD44 mRNA were detected by gel electrophoresis in NB and tumor-adjacent tissues The expression level. Results nm23 mRNA was expressed in both study group and control group, but the expression of nm23 mRNA was lower in the study group than in the control group. The expression of nm23 mRNA in the study group was lower than that in the non-lymph node metastasis group. CD44 mRNA was also expressed in the study group and the control group, but the expression of the study group was significantly higher than that of the control group, and the expression of CD44 mRNA in the study group was higher than that in the non-lymph node metastasis group. Conclusion The expressions of nm23 mRNA and CD44 mRNA are closely related to the occurrence, development and metastasis of NB. The expression of nm23 mRNA inhibits the occurrence and metastasis of NB, while the expression of CD44 mRNA promotes the development and metastasis of NB. Using PCR technology can effectively detect the expression of NB nm23 mRNA and CD44 mRNA, help to determine the biological behavior.
