目的探讨外源性肝细胞生长因子(HGF)能否调控肝癌细胞HEPG2 p16基因的表达水平。方法以人肝癌细胞株HepG2为研究对象,根据培养基中HGF的浓度不同,实验分组为:对照组、实验1组和实验2组,采用流式细胞仪(FCM)检测HGF的肝癌细胞生长周期;采用逆转录聚合酶链反应(RT-PCR)技术检测HGF对肝癌细胞p16基因表达的影响。结果HepG2细胞在10μg/L和50μg/L HGF的作用下,细胞周期出现明显改变,其中G0/G1期细胞明显多于对照组,而S期细胞减少。经HGF作用后肝癌细胞的p16基因表达上调。结论HGF通过上调p16表达阻滞细胞分裂于G0/G1期,可能为其抑制人肝癌细胞HepG2的生长作用的重要机制之一。
Objective To investigate whether exogenous hepatocyte growth factor (HGF) regulates the expression of HEPG2 p16 gene in hepatocellular carcinoma cells. Methods The human hepatocellular carcinoma cell line HepG2 was used as the research object. According to the concentration of HGF in the culture medium, the experimental groups were divided into control group, experiment 1 group and experiment 2 group. The growth cycle of HGF cells was detected by flow cytometry (FCM) The effect of HGF on the expression of p16 gene in hepatocellular carcinoma cells was detected by reverse transcription-polymerase chain reaction (RT-PCR). Results The cell cycle of HepG2 cells was significantly changed under the action of 10μg / L and 50μg / L HGF. The number of cells in G0 / G1 phase was significantly more than that in control group, while the number of S phase cells was decreased. After HGF treatment, the expression of p16 gene in hepatocellular carcinoma cells was up-regulated. Conclusion HGF up-regulates the cell division in G0 / G1 phase by blocking the expression of p16, which may be one of the important mechanisms of its inhibition on the growth of HepG2 cells.
