目的研究保健食品总黄酮含量测定的超声提取最佳条件。方法采用单因素分析法和正交试验法,以保健食品总黄酮含量为考察指标,研究不同料液比(1:25~1:5)、超声提取时间(10~40min)和乙醇浓度(50%~100%)对保健食品总黄酮提取量的影响。采用紫外分光光度法进行测定。结果由正交试验数据分析可知,影响因素的主次顺序是料液比>乙醇浓度>提取时间。在60%乙醇,料液比1:20,超声提取时间为20min条件下从保健食品中提取出的总黄酮含量最高,平均值为5.370mg。总黄酮的线性范围在1.0~40μg/ml之间,所得回归方程为y=0.0033x-0.0109,相关系数为0.9995,检出限为1.0μg/ml。该方法的加标回收率为95%~104%(n=3),RSD为1.717%(n=5)。结论该方法在最佳提取条件下进行,精密度和准确度均较高,空白低且稳定,适用于保健食品和某些食品中总黄酮的检测。
Objective To study the optimum conditions for ultrasonic extraction of total flavonoids in health food. Methods The content of total flavonoids in health food was investigated by single factor analysis and orthogonal test. The effects of different material-liquid ratio (1: 25-1: 5), ultrasonic extraction time (10-40 min) and ethanol concentration % ~ 100%) on the total flavonoids extract of health food. Uses the ultraviolet spectrophotometry to carry on the determination. Results Orthogonal test data analysis shows that the order of influencing factors is the ratio of solid to liquid> ethanol concentration> extraction time. The content of total flavonoids extracted from health food was the highest with the average ratio of 5.370mg in 60% ethanol, solid-liquid ratio 1:20 and ultrasonic extraction time 20min. The linear range of total flavonoids was 1.0 ~ 40μg / ml, the regression equation was y = 0.0033x-0.0109, the correlation coefficient was 0.9995, the detection limit was 1.0μg / ml. The recovery of this method was 95% ~ 104% (n = 3), RSD was 1.717% (n = 5). Conclusion The method was carried out under the best extraction conditions with high precision and accuracy, low blank and stable, suitable for the determination of total flavonoids in health food and some foods.
