色谱柱键合量是对色谱柱进行表征的一个重要参数,而现有文献对于蛋白质色谱柱的键合量测定至今未见报道。首次建立了利用考马斯亮蓝方法测定蛋白质色谱柱键载量的一种分析方法。同时考察了最大吸收波长、酸度、考马斯亮蓝浓度、测定时间等对测定蛋白质键载量的影响。采用物理吸附和化学键合2种方法制备了2种牛血清白蛋白色谱柱,测定了其蛋白质键合量。
The amount of column binding is an important parameter for the characterization of a chromatographic column. However, there is no report on the binding amount of a protein column in the existing literature. For the first time, an analytical method using Coomassie brilliant blue to determine the protein column loading was developed. The influence of the maximum absorption wavelength, acidity, Coomassie Brilliant Blue concentration, determination time and so on on the key protein loading was also investigated. Two kinds of bovine serum albumin (BSA) columns were prepared by physical adsorption and chemical bonding. The amount of protein binding was measured.
