目的 研究Fas/FasL在大肠癌及癌周不同部位细胞中的表达以及 p2 7、PCNA在大肠癌组织中的表达 ,并探讨其生物学意义。方法 利用免疫组化法对 50例大肠癌组织、癌旁交界组织、距肿瘤边缘 5cm处、正常组织的Fas/FasL表达进行检测 ,检验上述组织中Fas/FasL表达的差异。并对其中正常组织和肿瘤组织的 p2 7、PCNA在表达进行检测。 结果 比较肿瘤组织、癌旁交界组织与正常组织Fas的阳性表达率 (62 %vs84%vs1 0 0 % )和FasL的阳性表达率 (86 %vs66 %vs2 % ) ,差异均有显著性 (P <0 .0 5)。距肿瘤边缘5cm处与正常组织的Fas阳性表达率 (98%vs1 0 0 % )和FasL的阳性表达率 (8%vs2 % ) ,差异无显著性 (P >0 .0 5)。而不同分化的肿瘤组织中Fas/FasL表达差异无显著性 (P >0 .0 5)。p2 7在肿瘤组织与正常组织中的高表达率分别为 54 %和 1 0 0 % ;PCNA的阳性细胞数分别为 (2 2 .37± 6 .0 5)和 (50 .91± 1 6 .0 6) ,PCNA和p2 7在二组织中的表达差异均有显著性 (P <0 .0 5) ,且 p2 7、PCNA表达与大肠癌的分化程度、淋巴结转移和Dukes分期显著有关。结论 大肠癌中存在Fas表达的下调和FasL表达的上调 ,Fas/FasL系统可能参与了大肠癌细胞的免疫逃避机制 ;p2 7蛋白的表达与大肠癌的临床病理学特征密切相关 ,可能是一个?
Objective To study the expression of Fas / FasL in different parts of colorectal cancer and its adjacent tissues, and the expression of p27 and PCNA in colorectal carcinoma and its biological significance. Methods Immunohistochemistry was used to detect Fas / FasL expression in 50 cases of colorectal cancer tissues, paraneoplastic adjacent tissues, 5 cm away from tumor margin and normal tissues. The differences of Fas / FasL expression in the above tissues were examined. The expression of p27 and PCNA in normal tissues and tumor tissues were detected. Results The positive rates of Fas (62% vs 84% vs 100%) and FasL (86% vs 66% vs 2%) were significantly higher in tumor tissues, paraneoplastic junctional tissues and normal tissues (P < 0 .0 5). There was no significant difference in Fas positive expression rate (98% vs 100%) and FasL (8% vs2%) at 5cm from the edge of the tumor and normal tissue (P> 0.05). There was no significant difference in the expression of Fas / FasL in different differentiated tumor tissues (P> 0.05). The positive rates of p27 in tumor tissues and normal tissues were 54% and 100%, respectively. The numbers of PCNA positive cells were (22.37 ± 6.05) and (50.91 ± 1.6). There was significant difference in the expression of PCNA and p27 among the two tissues (P <0.05). The expression of p27 and PCNA was significantly correlated with the differentiation degree, lymph node metastasis and Dukes stage in colorectal cancer. Conclusion The down-regulation of Fas expression and the up-regulation of FasL expression in colorectal cancer may play an important role in the immune evasion mechanism of colorectal cancer cells. The expression of p27 protein is closely related to the clinicopathological features of colorectal cancer.
